The results showed that there is no significant difference in transmission ratio, Xist and X-linked genes expression, and Xist RNA localization between the female wild type (WT) and Xist P1-/- rabbits, suggesting that P1 is non-essential for Xist expression and XCI in rabbits. Our study has explored the function of Xist P1 in animal level for the first time, and the results provide new ideas for future studies of XCI mechanisms.Conflict adaptation reflects the increase in cognitive control after previous conflict between task-relevant and task-irrelevant information. Tonic (sustained) arousal elicited by emotional words embedded in a conflict task has previously been shown to increase conflict adaptation. However, the role of phasic (transient) emotional arousal remains unclear. In Experiment 1 (N = 55), we therefore investigated the effect of phasic arousal using a colour flanker task with negative, positive, and neutral words as stimuli. AZD8055 We hypothesised that phasic arousal elicited in this context will increase conflict adaptation in the subsequent trial. Indeed, when the words were positive or negative as compared to neutral, we observed increased conflict adaptation. We expected that emotional words with high self-relevance would lead to stronger effects of emotional arousal on conflict adaptation. Taken together, these results are the first to show that phasic arousal elicited by emotional words increases conflict adaptation, in particular when these words have high self-relevance.Characterizing bacterial communities is of great significance for targeted control of bacteria-induced clogging during geothermal water recharge. Based on a series of laboratory-scale percolation experiments, the variations in bacterial community diversity, composition, and structure were investigated during simulated geothermal water recharge using high-throughput sequencing technology. The Chao, Shannon, and Evenness indexes were used to quantify the richness, diversity, and evenness of the bacterial community, respectively. The results show that the richness of the bacterial community initially increased and then decreased in the sand columns during the experiments of geothermal water recharge, while the changes in bacterial diversity and evenness were not apparent. A variety of bacterial phyla were found, among which Proteobacteria was predominant (88.31%), followed by Actinobacteria, Bacteroidetes, and Firmicutes (4.23%, 3.44%, and 2.49%). For the non-Proteobacterial phyla, Actinobacteria gradually disappeared while Bacteroidetes and Firmicutes were detected during the percolation experiments. This study implies that, despite the variations in the bacterial community, a core group of bacteria persists during geothermal water recharge, and thus a targeted control of bacteria-induced clogging during geothermal water recharge should be feasible.This study aimed to screen putative drug targets associated with biofilm formation of multidrug-resistant Acinetobacter baumannii and Pseudomonas areugenosa and prioritize carbon nano-fullerene as potential lead molecule by structure-based virtual screening. Based on the functional role, 36 and 83 genes that are involved in biofilm formation of A. baumannii and P. areugenosa respectively were selected and metabolic network was computationally constructed. The genes that lack three-dimensional structures were predicted and validated. Carbon nano-fullerene selected as lead molecule and their drug-likeliness and pharmacokinetics properties were computationally predicted. The binding potential of carbon nano-fullerene toward selected drug targets was modeled and compared with the binding of conventional drugs, doripenem, and polymyxin-B with their usual targets. The stabilities of four best-docked complexes were confirmed by molecular dynamic (MD) simulation. This study suggested that selected genes demonstrated relevant interactions in the constructed metabolic pathways. Carbon fullerene exhibited significant binding abilities to most of the prioritized targets in comparison with the binding of last-resort antibiotics and their usual target. The four best ligand-receptor interactions predicted by molecular docking revealed that stability throughout MD simulation. Notably, carbon fullerene exhibited profound binding with outer membrane protein (OmpA) and ribonuclease-HII (rnhB) of A. baumannii and 2-heptyl-4(1H)-quinolone synthase (pqsBC) and chemotaxis protein (wspA) of P. aeruginosa. Thus, the current study suggested that carbon fullerene was probably used as potential lead molecules toward selected targets of A. baumannii and P. aeruginosa and the applied aspects probably scaled up to design promising lead molecules toward these pathogens.Communicated by Ramaswamy H. Sarma.In methodological and practical debates about replications in science, it is (often implicitly) assumed that replications will affect public trust in science. In this preregistered experiment (N = 484), we varied (a) whether a replication attempt was successful or not and (b) whether the replication was authored by the same, or another lab. Results showed that ratings of study credibility (e.g. evidence strength, ηP2 = .15) and researcher trustworthiness (e.g. expertise, ηP2 = .15) were rated higher upon learning of replication success, and lower in case of replication failure. The replication's author did not make a meaningful difference. Prior beliefs acted as covariate for ratings of credibility, but not trustworthiness, while epistemic beliefs regarding the certainty of knowledge were a covariate to both. Hence, laypeople seem to notice that successfully replicated results entail higher epistemic significance, while possibly not taking into account that replications should be conducted by other labs.
