Small interfering RNA was used to evaluate the role of Klotho in nicotinamide-related renoprotection. The results showed that nicotinamide attenuated renal pathologic morphology, kidney functional abnormalities, and kidney inflammatory response in rhabdomyolysis. In this study, we investigated the role of miR-383-5p in human MM cells in vitro. miR-383-5p expression was downregulated in MM cell lines compared with the human normal melanocyte cell line, and miR-383-5p overexpression inhibited the proliferation, migration, and invasion of M14 and A375 cells. Furthermore, miR-383-5p was able to effectively bind to the 3'UTR of CENPF mRNA. miR-383-5p expression was negatively correlated with CENPF expression and miR-383-5p overexpression inhibited CENPF protein expression in M14 and A375 cells. The overexpression of CENPF could effectively rescue the inhibitory effect on proliferation and invasion caused by miR-383-5p. Additionally, using publicly available databases, we showed that CENPF expression was upregulated in human MM tissues and could predict the prognosis of MM. However, their roles in non-alcoholic fatty liver disease (NAFLD) of laying hens have not been reported. Here, we investigated the effects of different concentrations of GEN and BPA on the NAFLD of laying hens. Results showed that GEN ameliorated the high-energy and low-protein diet (HELP)-induced NAFLD by improving pathological damage, hepatic steatosis, and insulin resistance and blocking the expression of NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome-related factors. By contrast, high dose of BPA could aggravate these changes with serious symptom of NAFLD and suppress the level of ERα in the liver considerably, while GEN could reverse this phenomenon in a dose-dependent manner. In general, our research shows that the protective effect of GEN on NAFLD aims to improve the metabolic disorders and inflammation closely connected to ERα, while BPA can inhibit the expression of ERα and exacerbate the symptom of NAFLD. In conclusion, we elucidate the opposing effects of GEN and BPA in NAFLD of laying hens, thus providing a potential mechanism related to ERα and inflammation.There is currently a significant gap in knowledge about the emission and deposition of mercury (Hg) from coal-fired power plants in Australia. To fill this gap, we propose a novel method that combines several sources of information (stratigraphic data, hydrodynamic modelling and atmospheric modelling), to identify the sources and fates of Hg emitted from coal-fired power plants. The stratigraphic record from Lake Macquarie (Australia) shows that mercury deposition increased up to 7-times since the 1950s, which is when coal-fired power plants were commissioned in the catchment. The stratigraphy also shows a decrease in Hg deposition with power plant retrofits. Using results from multiple models (statistical modelling, hydrodynamic modelling, particle density modelling and atmospheric emissions modelling), we found that ash dams contribute little Hg to Lake Macquarie. Instead, most of the Hg contamination in the lake is a result of atmospheric emissions from the power plants, and these power plants are also depositing Hg in the urban areas to the west of the lake. Our results demonstrate that the multi-proxy approach demonstrated in the paper can be used to provide clues as to the source of Hg, so that appropriate mitigation strategies and regulatory frameworks can be implemented.
