Pre- and post-treatment tumour biopsies for two patients identified temporal genomic heterogeneity, somatic mutations in the gene, which encodes a PI3K-like protein kinase, and emergent somatic mutations related to protein kinase signalling. Copanlisib and trastuzumab can be safely administered with fair overall tolerability. Preliminary evidence of tumour stability was observed in patients with heavily pre-treated, metastatic HER2 positive breast cancer. Several potential biomarkers were identified for further study in the current phase 2 clinical trial. New solutions are sought, firstly, to reduce heat losses and, secondly, to improve the environment conditions in isolated rooms. The effective heat resistance of insulating materials is inversely proportional to temperature and humidity. Cement composites filled with lightweight artificial aggregates may be a suitable material. Selecting a proper method for measuring the thermal conductivity of concrete is important to achieve accurate values for calculating the energy consumption of buildings. The steady state and transient methods are considered the two main thermal conductivity measurement approaches. Steady state is a constant heat transfer, whereby the temperature or heat flow is time independent. In the transient method, temperature changes over time. Most researchers have measured the conductivity of cement-based materials based on transient methods. The availability and cost of equipment, time for experimental measurements and measurement ability for moist specimens may be some of the reasons for using this method. However, considering the accuracy of the measurements, the steady state methods are more reliable, especially for testing dry materials. Four types of composites were investigated that differed in filler natural aggregate, sintered fly ash filler, sintered clay and granular foam glass aggregate. The method of preparing the samples for testing is especially important for the obtained results. The samples, with a specific surface roughness, will show a lower coefficient of thermal conductivity by 20-30%; therefore, the selection of the type of contact layer between the plate of the measuring device and the sample is of particular importance.Measurement of ice nucleation (IN) temperature of liquid solutions at sub-ambient temperatures has applications in atmospheric, water quality, food storage, protein crystallography and pharmaceutical sciences. Here we present details on the construction of a temperature-controlled microfluidic platform with multiple individually addressable temperature zones and on-chip temperature sensors for high-throughput IN studies in droplets. We developed, for the first time, automated droplet freezing detection methods in a microfluidic device, using a deep neural network (DNN) and a polarized optical method based on intensity thresholding to classify droplets without manual counting. This platform has potential applications in continuous monitoring of liquid samples consisting of aerosols to quantify their IN behavior, or in checking for contaminants in pure water. A case study of the two detection methods was performed using Snomax® (Snomax International, Englewood, CO, USA), an ideal ice nucleating particle (INP). Effects of aging and heat treatment of Snomax® were studied with Fourier transform infrared (FTIR) spectroscopy and a microfluidic platform to correlate secondary structure change of the IN protein in Snomax® to IN temperature. It was found that aging at room temperature had a mild impact on the ice nucleation ability but heat treatment at 95 °C had a more pronounced effect by reducing the ice nucleation onset temperature by more than 7 °C and flattening the overall frozen fraction curve. Results also demonstrated that our setup can generate droplets at a rate of about 1500/min and requires minimal human intervention for DNN classification.Importance The high incidence of major depressive episodes during interferon-α (IFN-α) therapy is considered the most powerful supportive evidence for the inflammation theory of depression. As the kynurenine pathway plays an important role connecting inflammation and depression, it is plausible to investigate this pathway for predictive genetic markers for IFN-α-induced depression. Methods In this prospective case-control study, we assessed 291 patients with chronic hepatitis C viral infection taking IFN-α therapy and analyzed the single nucleotide polymorphisms (SNPs) in genes in the kynurenine pathway. Our case group contained patients who developed IFN-α-induced depression during the treatment, and others were defined as the control group. Genomic DNA was extracted from leukocytes in the peripheral blood and analyzed by Affymetrix TWB array. We first tested allelic, dominant, and recessive models on each of our SNPs using Fisher's exact test. We then conducted 5000 gene-wide max(T) permutations based on thrtive evidence of the involvement of the kynurenine pathway in IFN-α-induced depression. SNPs in this pathway were also predictive of this disease. The interesting properties of Al 6061 aluminum foams have boosted the research on the correlation between foam composition and morphology and its mechanical response under dynamic conditions. In this study, ingots of an Al 6061-T4 foam were sectioned and analyzed in order to determine their microstructural and morphological characteristics, and then quasi-static and dynamic tests (10-3 to 3 × 102 s-1) were carried out to determine the material mechanical behavior. Dynamic tests, carried out by using the split Hopkinson bar, highlighted that the studied foam is characterized by a very good energy absorption capability, due to its ductile behavior. Nevertheless, the conducted research showed that cell morphology and distribution affect its mechanical behavior in dynamic conditions in which localized cell collapse may result in a decreased energy absorption and efficiency of the foam.
