Pregabalin (PGL) is a gabapentinoid used to treat epilepsy, neuropathic pain and generalized anxiety disorder. PGL is also misused by heroin users as it enhances the effects of heroin. While it is thought those who misuse PGL take it in amounts greater than the recommended therapeutic dose, it is unknown whether there is a significant difference between the amounts of PGL used by heroin users compared to non-heroin users. This study hypothesised that the PGL concentrations in post-mortem samples taken from heroin users positive for PGL would be higher than those in non-heroin users. Between 01.01.2016 and 31.12.2017, a routine drugs screen and a specific screen for PGL was carried out on femoral-vein bloods from 3,750 post-mortem Coroners' cases. Of the cases screened, 354 were heroin users, of which 264 cases were negative for gabapentinoids and therefore used as the control-heroin users group. PGL was positive in 229 cases, of which 69 were heroin users and 160 were non-heroin users. On comparing the PGL cooxicity and death in this population.Plant nitrogen acquisition requires carbon to be allocated belowground to build roots and sustain microbial associations. This carbon cost to acquire nitrogen varies by nitrogen acquisition strategy; however, the degree to which these costs vary due to nitrogen availability or demand has not been well tested under controlled conditions. We grew a species capable of forming associations with nitrogen-fixing bacteria (Glycine max) and a species not capable of forming such associations (Gossypium hirsutum) under four soil nitrogen levels to manipulate nitrogen availability and four light levels to manipulate nitrogen demand in a full-factorial greenhouse experiment. We quantified carbon costs to acquire nitrogen as the ratio of total root carbon to whole-plant nitrogen within each treatment combination. In both species, light availability increased carbon costs due to a larger increase in root carbon than whole-plant nitrogen, while nitrogen fertilization generally decreased carbon costs due to a larger increase in whole-plant nitrogen than root carbon. Nodulation data indicated that G. max shifted relative carbon allocation from nitrogen fixation to direct uptake with increased nitrogen fertilization. These findings suggest that carbon costs to acquire nitrogen are modified by changes in light and nitrogen availability in species with and without associations with nitrogen-fixing bacteria.Chlorine is a toxic chemical that has been used as a chemical warfare agent in recent armed conflicts. There is an urgent need for methods to verify alleged uses of chlorine, and phospholipid chlorohydrins (PL-HOCl) derived from the pulmonary surfactant of exposed victims have previously been proposed as biomarkers of chlorine exposure. Here we describe an improved protocol for the chemical analysis of these biomarkers and its applicability to biomedical samples from chlorine-exposed animals. By the use of a polymeric solid phase-supported transesterification of PL-HOCl using ethanolamine, a common biomarker; oleoyl ethanolamide chlorohydrin (OEA-HOCl), was derived from all the diverse oleoyl PL-HOCl that may be formed by chlorine exposure. GSK3368715 mouse Compared to native lipid biomarkers, OEA-HOCl represents a larger biomarker pool and is better suited for nano-liquid chromatography tandem mass spectrometry (nLC-MS/MS analysis), generating 3 amol LOD and a reduced sample carry-over. With the improved protocol, significantly elevated levels of OEA-HOCl was identified in broncho-alveolar lavage fluid (BALF) of chlorine exposed rats, 2-48 hours after exposure. The difficulty of BALF sampling from humans limits the methods usefulness as a verification tool of chlorine exposure. Conversely, nasal lavage fluid (NLF) is readily collected without advanced equipment. In NLF from chlorine-exposed rats, PL-HOCl were identified and significantly elevated levels of the OEA-HOCl biomarker was detected 2- 24 hours after exposure. In order to test the potential of NLF as a biomedical sample for verification of human exposure to chlorine, in-vitro chlorination of human NLF samples was performed. All human in-vitro chlorinated NLF samples exhibited elevated OEA-HOCl biomarker levels, following sample derivatization. This data indicates the potential of human NLF as a biomedical sample for the verification of chlorine exposure but further work is required to develop and validate the method for the use on real-world samples.Bioadhesives are wet or dry polymeric compounds that rely upon physical and chemical properties to generate characteristic sticky forces. The past decade has seen a rapidly evolving field of research around the functions, genetics, biochemistry, and mimetics of bioadhesives, but challenges unique to this research area continue to arise. We polled the presenters of SICB Symposium #8 to describe the "Ties that Stick" challenges and exciting prospects that most resonated with their research pursuits. Themes that emerged from the poll included difficulties working with adhesive-producing organisms, field inherent knowledge gaps in theoretical modeling, molecular interactions, technology, and the interdisciplinary rigor of the bioadhesives field. We address each challenge with a discussion of the opportunities and applications presented by bioadhesives research. To investigate the longitudinal associations of serum inflammatory markers and adipokines with joint symptoms and structures in participants with knee osteoarthritis (OA). Two hundred participants (46.5% female, mean age 63.1 years, mean BMI 29.5 kg/m2) from Tasmania part of the VIDEO (Vitamin D Effect on OA) study were randomly selected in the current study. Serum levels of 19 biomarkers, scores of Western Ontario and McMaster Universities Index (WOMAC), and magnetic resonance imaging-assessed knee structures were evaluated at baseline and month 24. The patterns of biomarkers were derived from principal component analysis and their association with knee symptoms and structures were examined using adjusted generalized estimating equations. Five components explained 78% of the total variance. Interleukin (IL)-1β, -2, -4, -6, -8, -17A, -17F, -21, -22 and -23 loaded the highest on the first component, which was associated with increased bone marrow lesions (BMLs) and WOMAC dysfunction score. IL-10, -12 and granulocyte-macrophage colony-stimulating factor loaded on the second component, which was associated with increased cartilage volume, and decreased effusion-synovitis and WOMAC scores.
