Coffee residue extracts were extracted from coffee residue, produced after coffee extraction for coffee powder and instant coffee preparation, using an autoclave. Leuconostoc mesenteroides WiKim32 was selected to evaluate the ability of coffee residue extracts to protect bacteria during freeze-dried storage. The storage stability of freeze-dried Leu. mesenteroides WiKim32 with coffee residue extracts was comparable to those with commercial cryoprotective agents. Coffee residue extracts contributed to storage stability immediately after freeze-drying (61.2%) and subsequent storage (48.7%). Our data indicate that the protective effect of the coffee residue extracts is associated with ions, carbohydrates, and phenolic compounds. Coffee residue extracts are feasible materials, which can reduce the storage and distribution costs compared to commercial agents currently available.Hutchinson-Gilford Progeria Syndrome (HGPS) is a rare autosomal, dominant genetic condition characterized by many features of accelerated aging. On average, children with HGPS live to about fourteen years of age. The syndrome is commonly caused by a point mutation in the LMNA gene which normally codes for lamin A and its splice variant lamin C, components of the nuclear lamina. The LMNA mutation alters splicing, leading to production of a truncated, farnesylated form of lamin A referred to as "progerin." Progerin is also expressed at very low levels in healthy individuals and appears to play a role in normal aging. HGPS is associated with an accumulation of genomic DNA double-strand breaks (DSBs), suggesting corruption of DNA repair. In this work, we investigated the influence of progerin expression on DSB repair in the human genome at the nucleotide level. We used a model system that involves a reporter DNA substrate inserted in the genome of cultured human cells. A DSB could be induced within the substrate through exogenous expression of endonuclease I-SceI, and DSB repair events occurring via either homologous recombination (HR) or nonhomologous end-joining (NHEJ) were recoverable. Additionally, spontaneous HR events were recoverable in the absence of artificial DSB induction. We compared DSB repair and spontaneous HR in cells overexpressing progerin versus cells expressing no progerin. We report that overexpression of progerin correlated with an increase in DSB repair via NHEJ relative to HR, as well as an increased fraction of HR events occurring via gene conversion. Progerin also engendered an apparent increase in spontaneous HR events, with a highly significant shift toward gene conversion events, and an increase in DNA amplification events. Such influences of progerin on DNA transactions may impact genome stability and contribute to aging. To account for hip contribution, a double inverted pendulum (DIP) model rotating about the ankle and hip joints has been recently proposed in our previous work but experimental validation efforts are scarce. In the present study, it is investigated whether the DIP model is able to reproduce the experimentally observed frequency spectrum of the ankle and hip joint kinematic for young and elderly subjects balancing on a compliant surface. see more The DIP model based and experimental kinematics are compared via Fourier analysis to obtain their corresponding amplitude spectrum density (ASD) functions. Quantitative comparisons of the ASD functions are accomplished through Bland-Altman (B&A) analysis, and Peger than 0.35 Hz. Moreover, the model captures the decaying behavior of the experimental ASD functions as frequency increases. With respect to joint angle velocities, the highest PCC between model-based and experimental ASD functions is found for the hip joint of elderly subjects. The B&A analysis shows that the zero-difference between model-based and experimental ASD functions lies between the 95 % confidence interval, especially for the joint angle position results. These suggest that the DIP model reproduces part of the experimentally observed frequency spectrum, which validates the model to study the dynamics and stability of the human upright posture. Previous studies based on voxel-based morphometry (VBM) had revealed brain gray matter (GM) changes in chronic smokers relative to nonsmokers. However, not all studies reported entirely consistent findings, or even opposite. The aim of this study was to conduct a quantitative meta-analysis of VBM studies of chronic smokers. A systematic database search was conducted in PubMed and Web of Knowledge from January 1, 2000 to January 31, 2020 to identify eligible VBM studies. Meta-analysis was performed with the Seed-based d Mapping software package to compare alternations between chronic cigarette smokers and nonsmokers. In addition, meta-regression analysis were performed to examine the influences of cigarette per day, smoking history and FTND. A total of 17 VBM studies including 905 smokers and 1344 nonsmokers met the inclusion criteria. The results of this meta-analysis showed that the chronic smokers showed a robust GM volume decrease in bilateral prefrontal cortex and left insular and a GM increase in the right lingual cortex and left occipital cortex. Moreover, meta-regression analysis showed that cigarette per day, smoking history and FTND were partly associated with GM changes in chronic smokers. This meta-analysis indicated that chronic cigarette smokers had significant and robust brain GM alternations compared with nonsmokers.
