Shui people's healthcare has been greatly influenced by their accumulated and refined traditional medicinal knowledge. Rural development in Guizhou Province relies heavily on traditional ethnic herbal medicines, with Shui ethnomedicine playing a key role. However, a paucity of investigation has been undertaken on the medicinal plants customarily employed by the Shui people. The current study's objectives include (1) documenting the present use of medicinal plants in Sandu County, encompassing associated traditional knowledge, including details of plant species used, diseases treated, and unique preparation methods employed by local healers; (2) performing a quantitative analysis of prominent medicinal plant species through relative frequency of citation; and (3) providing valuable data for prospective ethnomedicine advancements and applications, and promoting the preservation of Shui traditional medicinal knowledge.The period of field surveys in Sandu County encompassed July 2015 to August 2022. A total of fifteen local healers, identified as key informants, were interviewed for the study. Further interviews, employing semi-structured methodologies and focal group discussions, were conducted with 132 additional informants from villages and local markets. In the fields, Shui healers, while gathering medicinal plants, were observed and their plant selections, uses, and collected parts meticulously recorded. An ANOVA analysis was performed to investigate the number of medicinal plants recognized by local healers in different age groups and townships, along with calculating the relative frequencies of the cited plants.This study involved the analysis of data obtained from 15 Shui healers and 132 additional informants. For treating a vast variety of conditions and diseases, healers used the fresh or dried parts of a total of 505 plant species. Of the 156 families and 405 genera encompassing these plants, Fabaceae stands out as the most prevalent family. The concentration of per capita healers (pch) reached its highest level in Jiuqian township, significantly contrasted by Zhonghe's singular local healer. Among the fifteen local healers, a mere two individuals were under forty years old. A significant worry exists regarding the potential loss of traditional medicinal knowledge in the absence of adequate apprenticeships or suitable inheritors. The Shui often employ the entire plant, given that herbaceous plants are common among medicinal species. Among the various diseases treatable by Shui medicinal plants, rheumatism and bone fractures are associated with the largest number of different plant species utilized in their treatment. From the peripheral blood of every patient, a whole-genome capture of the DMD gene was extracted and next-generation sequencing was executed. MLPA analysis verified large copy number variants (CNVs) within the exon regions of the DMD gene, and Sanger sequencing corroborated smaller variations like single nucleotide variations and insertions/deletions of fragments fewer than 50 base pairs in length.A noteworthy 96.1% (49/51) of the 51 patients possessed pathogenic or likely pathogenic variations in the DMD gene. From the 49 positive specimens, 17 patients (comprising 347% [17 out of 49]) showcased CNVs in exonic segments, with 32 patients (accounting for 653% [32/49]) exhibiting small variations. Among the identified novel variants, we found c.10916_10917del, c.1790T>A, c.1842del, c.5015del, c.5791_5792insCA, and the duplication of exons 38 through 50.A study of 51 patients revealed pathogenic or likely pathogenic DMD gene variants in 49 patients (96.1% of the total). A noteworthy 6 variants (12.2% of the affected patients) were found to be previously unreported.
