Finally, a general proposal is presented on the conceptual design of automation-optimized computational image analysis workflows in the biomedical and clinical domain.When sampling an item or surface for DNA, the collection of 'background' DNA (bDNA) from previous use poses an issue as it may impact the detectability of 'target' DNA and the interpretation of the DNA results given alleged activities. This study investigates the prevalence and transferability of bDNA on flooring surfaces within occupied houses under conditions similar to those that are encountered in casework. To assess bDNA presence and transferability, and the impact of how and who contacts the surface, areas used frequently and infrequently were targeted in the kitchen, living room, bedroom and bathroom of five houses, and two samples taken from each area; one directly from the floor and another from a cotton surface after contacting the floor. DNA was detected in 97 % (of 39) of samples collected directly from flooring, with 92 % providing interpretable profiles. DNA was detected in 85 % (of 39) samples collected from cotton swatches after contacting the floors, with 79 % providing interpretable profiles they were known to use and could be observed in profiles collected from rooms they reportedly did not use. The findings of this preliminary investigation provide an understanding of the complexities of transfer, persistence, prevalence and recovery of DNA traces in houses occupied by multiple people and highlights the need to consider how and who uses a space, in the investigation of criminal activities where DNA traces are recovered from, or have been in contact with, flooring.The goal of this study was to characterize the reproducibility of extracellular and cell pellet associated DNA yields recovered from handled substrates. Results showed that extracellular DNA yields were extremely variable between contributors-ranging between 0 and >10ng-and tended to dwarf cell pellet yields, which varied between 0 and ∼230pg. DNA yields across multiple samples from the same contributor on different days showed similar levels of variability in both DNA fractions, indicating that extracellular DNA yield is largely influenced by extrinsic and/or environmental factors and is not a contributor-specific attribute. In this work, a universal GC-MS method has been developed to analyze various ink components extracted from inkjet printouts. The results indicated that several components detected and identified across 195 inks could be used to distinguish printer manufacturers. A trend of decreasing solvent concentration over time was observed through the continuous monitoring of 7 samples. The results shown that this method is useful for forensic classification purposes, and can be useful regardless effects of storage environment, paper or printer. Furthermore, the application of this method in the analysis of counterfeit banknotes illustrated its feasibility and applicability.Multi-isotope analysis (e.g., Sr-Pb-O-H-C-N) of human scalp hair is routinely used in forensic investigations of human remains to constrain the geographic origin of unidentified bodies, and to investigate antemortem mobility patterns. However, while it is known that postmortem processes can affect the preservation of, or even overprint, the biogenic isotopic signatures in hair, the speed and nature of these processes have rarely been studied. This study investigates the effects of decomposition and environment on the H-Pb-Sr isotope compositions of human hair as well as the relationship between structural hair shaft degradation and isotopic signature change over time. Human scalp hair samples from four body donations were collected at different stages throughout gross body decomposition. The willed-donated bodies were placed to decompose outdoors at the Forensic Anthropology Research Facility (FARF) at Texas State University. Hair fibres from two of the donations were examined using scanning electron microscopy (SEM) and high-resolution light microscopy (HRLM). Chemical and microbiological degradation of hair fibres occurred rapidly after placement of the body outdoors. Measurements of scalp hair isotopic composition demonstrated that H-Pb-Sr isotope ratios were altered within days after environmental exposure, presumably by deposition, leaching and/or exchange with the local bioavailable soil, and vapour. The degree of physical hair degradation and changes in H-Pb-Sr isotope composition were not correlated. We conclude that antemortem isotopic H-Pb-Sr isotope ratios are difficult to recover in hairs derived from decomposing whole bodies. The purpose of this study is to investigate whether the use of suicidal asphyxiation by means of the helium method substituted other, related, methods following its publication in the Netherlands. We analysed suicides in the Netherlands over the period from 1 July 2012 to 30 June 2019. We compared the number of deaths caused by the helium method with other, related, cases. Secondly, we related these deaths to the total number of inhabitants and suicides recorded by Statistics Netherlands. The study showed a stable trend in the use of the helium method in the period 2012-2019 and this was the same for the other, related methods. Individuals using the helium method were significantly younger than those using other, related, methods.
