BACKGROUND Cubilin (CUBN) gene was reported to be a novel risk variant for colorectal cancer (CRC). see more Previous studies have shown that germline variants in known cancer driver genes are predictive of patient outcome, but no study has systematically analyzed CRC to identify CUBN that can predict patient outcome and function by using bioinformatics. MATERIAL AND METHODS The association in expression, clinicopathological parameters, and survival were analyzed by using Oncomine, UNCLA, and GEPIA, while CUBN alterations and related functional networks were identified using cBioPortal. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes pathways (KEGG) of CUBN in CRC were explored by using LinkOmics. Gene set enrichment analysis (GSEA) examined target networks of kinases, miRNAs, and transcription factors. RESULTS We found that CUBN was overexpressed in CRC. Patients who were in advanced TNM stage tended to express higher CUBN mRNA levels, while those who received radiotherapy tended to express relatively lower CUBN mRNA levels. Higher expression of CUBN was found to be associated with shorter overall survival (OS) and disease-free survival (DFS). Moreover, functional networks analysis suggested that CUBN can regulate mismatch repair, terpenoid backbone biosynthesis, base excision repair, and proteasome via vitamin digestion and absorption pathway to influence CRC occurrence. CONCLUSIONS These findings suggested that CUBN could serve as a prognostic and therapeutic biomarker of CRC in the future.BACKGROUND Successful endodontic therapy requires a fluid-tight apical seal. The aim of this in vitro study was to evaluate and compare the apical sealing ability of 3 root canal sealers Apexit Plus, AH Plus, and Resilon/Epiphany sealers. MATERIAL AND METHODS Samples of 152 single-rooted teeth were divided into 2 test groups, and each group was divided into 3 subgroups based on the preparation and obturation technique. After preparation with the step-back technique, the first group of root canals were obturated with lateral gutta-percha compaction in combination with AH Plus sealer or Apexit Plus or with the Resilon/Epiphany system alone. In the second group, preparation was done with the crown-down technique and root canals were then obturated with Thermafil obturator in combination with AH Plus or Apexit Plus or with Resilon/Epiphany sealer alone. Apical leakage was determined using a dye leakage test observed with a stereomicroscope. The degree of dye leakage was assessed using the t test to comparing the arithmetic averages of the groups. RESULTS In the groups prepared with the step-back technique, the average dye leakage was lower in samples obturated with Resilon/Epiphany, than in those filled with AH Plus/lateral gutta-percha compaction or Apexit Plus/lateral gutta-percha compaction. In groups prepared with the crown-down technique and obturated with the Resilon/Epiphany system, the average dye leakage was lower than in those filled with AH Plus/Thermafil obturation and those filled with Apexit Plus/Thermafil obturation. CONCLUSIONS Although all preparation and obturation techniques showed dye penetration, the crown-down technique paired with the Resilon/Epiphany system showed the least leakage. The way people derive inferences from actual adverse experiences plays an important role in the development of psychopathology. This study aims to examine the mediating role of pathogenic beliefs (i.e., emotion-laden, powerful, painful convictions about self and others) on the relation between perceived adverse parenting behaviors in childhood and subsequent adult psychopathology. Participants (mostly Caucasian and heterosexual) were 204 consecutively admitted patients with a range of psychological difficulties, including depression, anxiety, and interpersonal problems, at a low-fee outpatient clinic. Participants completed standard self-report assessments of perceived parental style, depressive and anxiety symptoms, and a clinically derived measure of pathogenic beliefs. We examined the indirect effects of adverse parenting on anxiety and depressive symptom severity through pathogenic beliefs. Pathogenic beliefs reflecting the unreliability of others significantly mediated the relationship between adverse our results seem to suggest that attending to intermediary subjective beliefs might be important in understanding psychopathology development in the context of childhood adversity. Aiming to modify the beliefs in therapy might modify the symptoms. However, this would remain to be demonstrated through formal intervention research. The impact of religiosity and spirituality (R/S) on mental health and the importance of incorporating R/S in mental health treatment have been previously explored. However, clarification is needed regarding the effects of R/S in the process of recovery from persistent mental disorders (PMDs). This study identifies how different R/S factors correlate with the stages of recovery, while considering individual psychological well-being and perceived social support. Seventy-five individuals who use Madrid's public services for people with PMD completed questionnaires assessing those factors. We found that during the recovery process, R/S correlated with psychological well-being and perceived social support, and that these variables were associated with the five stages of recovery differently. We further identified the importance of gratitude, compassion, inner peace, and connection with life in the recovery process. Overall, our findings highlight the importance of these R/S variables in particular recovery stage particular recovery stages for individuals with PMD. Acute focal demyelination is the characteristic feature of multiple sclerosis, with the majority of damaged axons undergoing limited remyelination and forming chronic lesions. Potential remyelinating agents are currently under development and there is therefore an urgent need for reliable in vivo biomarkers of remyelination. This study aimed to investigate potential changes in multifocal visual evoked potentials' (mfVEPs) latency in a cohort of relapsing-remitting multiple sclerosis (RRMS) patients. The potential sample size required for a remyelination-based clinical trial using different treatment effect sizes and the mfVEP latency as an outcome measure was also estimated. A total of 50 RRMS consecutive patients with no previous history of optic neuritis in at least one eye and 15 normal controls of similar age and gender composition were prospectively enrolled. Fifteen patients had a history of unilateral ON more than 12 months earlier, whereas 41 patients demonstrated optic radiations lesions on MRI at baseline.
